In the Marvel universe, while many superheroes are certainly mutated by radioactive spiders or cosmic rays (e.g., Spider-Man, the FF), the term "Mutant" is typically reserved for someone born with the X-Gene, which typically triggers around adolescence, usually causing sudden development of abnormal powers. The X-Gene marks them as homo sapiens superior - a new subspecies evolved from homo sapiens.
So while Reed and Sue Richards are not "mutants," their son, Franklin Richards was born a mutant because of their mutations and is among the most powerful in the Marvel Universe.
